This powerpoint presentation begins by stating the study motivation that includes the following: Current Regulatory Framework -
protection of public health from pathogens such as
Cryptosporidium (LT2), and
management of DBPs and organics (Stage 2 D/DBP Rule); and,
Chlorine Disinfection
- effective for many microorganisms under proper
conditions,
- some viruses (adenovirus) and protozoa (Cryptosporidium)
resistant to chlorination, and
- reacts with organic matter to form regulated DBPs. A brief overview of sonication is presented, along with a flow-through cavitation unit. Experimental objective: quantify E. coli inactivation, known concentration of organisms in chlorine demand free PBS, disinfection by sonication, chlorination, or both, and monitored E. concentration, power output, and temperature. - E. coli cultured in laboratory to stationary phase,
centrifuged and decanted,
- resuspended in phosphate buffered saline (PBS) at starting
concentration of ~107 cells/mL,
- enumerated by pour plates on tryptic soy agar. Preliminary results and related research are provided. Includes table, figure.
